【SFBC Lecture】Chemical Approaches to Sorting Out Protein Acetylation

Lecture Abstract: 

Lysine acetylation on histones and other proteins is critical to the regulation of gene expression and other cellular processes. We will describe our focus on the development of designed p300/CBP histone acetyltransferase (HAT) inhibitors as biological tools. We will also discuss aspects of p300/CBP HAT mechanism and regulation gleaned from enzymatic studies. In addition, methods for installing acetyl-Lys mimics and HAT inhibitor elements site-specifically into peptides and proteins will be highlighted.

 

Lecturer Profile: 

Philip Cole 教授目前担任美国约翰霍普金斯大学医学院药理学分子科学和药理学系系主任,Marshall-Maren 教授、生物活性分子中心主任以及悉尼金梅尔综合癌症中心化学治疗联合主任。作为表观遗传学和化员工物学领域的顶尖科学家,他长期从事蛋白质翻译后修饰的酶的机制和抑制研究。多年致力于发展一系列研究转录后修饰的化员工物学方法,如目前获得广泛应用的蛋白质工程方法“Expressed protein ligation”等,这些化员工物学方法极大地促进了表观遗传学研和蛋白质科学研究。他设计的GOAT、蛋白激酶、组蛋白乙酰基转移酶和组蛋白去甲基化酶特异抑制剂对化员工物学、蛋白激酶与信号传导、免疫学与代谢治疗等作出了重要贡献。 近年来,他在Science(3)、Cell(2)、Nature(4)、Nature 子刊(11)、Mol Cell(5)、PNAS(11)、JACS(15)等国际顶尖学术刊物发表了160 余篇高质量研究论文。基于其对化员工物学领域的杰出贡献,自80年代中期以来,Cole 教授荣获了一系列国际科技大奖如Irving A Hansen Memorial Foundation Award 和Ellison Medical Foundation New Scholar, 并曾经被评为Innovator of the Year,并担任多所国际著名大学和研究机构的lecture。
